Being it be your first time in Vegas. Do the whole casino hopping thing. Get to know what the strip and off strip casinos offer to see which you are comfortable with. That'll be a good thing to do for your first trip. Its hard to take it all in the first time in my opinion.

In thinking of what a group celebrating a 21st birthday might like... Hard Rock and/or Palms. Both are off the strip however, so while both "cater" to the type group you are, you'll find yourselves in cabs headed to the strip guaranteed... so why not stay there. Cosmopolitan is still the "in" spot on the strip for now. Younger energy and a very nice facility for partyin'. Planet Hollywood would be my 2nd recommendation on the strip for you. Great location, lots of stuff to do.
BTW... I also agree that Encore is the "best" hotel/casino in Las Vegas, but I've always found it to be a bit on the "stuffy" side and for a group celebrating a 21st? Ehhhhhh
I actually think the best "casino" in Vegas is the golden nugget or red rock ranch. This is from a gamblers perspective alone.
Golden Nugget has more table game variety and affordable tables. Most tables are $5-10 They have 2 deck blackjack at a $10 min (most other casinos are $50-$100 for this extremely low rake game) It's easy to earn comps and they don't nickel and dime you. This is a true gamblers old Vegas casino.
Red rock has some increased odds games like single zero roulette. And high paying slots.
Both are nice clean casinos. There are others that offer this but are super warn down.
The strip has some of the worst casinos in the world odds wise. But they're beautiful resorts. I'd check out bellagio Wynn/encore and cosmo. For resorts.
But it also depends on your game, for blackjack the hacienda famously offers a single deck, hit soft 17, double after split, no surrender, re-split aces table with a two dollar minimum. A house edge of only 0.02%. The best black jack in the world.
Roulette several casinos offer single 0 roulette on the strip. But planet Hollywood offers it at pretty low limits. 00 roulette 5.3% edge. Single 0 is only 2.7%

Sorry I re read that and felt like I sounded like a total bitch. If you want to have fun and party, try the hard rock, it's awesome, and planet Hollywood. Encore is great if you can afford it. Also if you have guys in your group, check out freemont st and more specifically freemont street east. It has a lot of really cool non casino bars. This is really what people picture as Vegas.
I remember one time I was playing blackjack at a $25 minimum table at Bellagio, slid my players club card to the dealer, who promptly slid it back at me and said "Why don't you give this to us when you start playing with real $$?". I burned through my remaining chips and left. And I'm a fancy old man no less! lol
A) It isn't very large; your group will be near each other the whole time.
B) It is usually about $5 minimum cheaper than the strip at any given time (meaning it is $10 minimum when the strip is at $15, etc).
There's nothing worse than hitting a casino (like say the bellagio or MGM) and your buddies all fan out to find a seat at a $15 table and you never play together.

TI also has pretty cheap tables and good dealers.
Wynn is my personal favorite and I like their rewards system better than Palazzo/Venetian. Wynn has a lot of young people gambling too especially at night.
Not a fan of Cosmo's floor set up and didnt enjoy it that much
