According to this link, Club Rio (in The Rio) was the first big club inside a hotel. That was 1995.

man, club rio and bikini beach bar at the rio; ra at luxor; rumjungle at mandalay bay; studio 54 at mgm; venus at venetian(!!). but i think it was really rain and ghostbar at the palms that upped the game on everyone and everywhere. those days were definitely different back then!!
